The Nanophore 5 genetic analyzer was produced by the Russian scientific production company Syntol in cooperation with the Institute of Analytical Instrumentation of the Russian Academy of Sciences (St. Petersburg) and the Experimental Plant of Scientific Instrumentation of the Russian Academy of Sciences (Chernogolovka). The employees of the Laboratory of Genetic Engineering note the accuracy and reliability of the measuring instrument.The Nanophore 5 genetic analyzer is used to determine the DNA sequence (sequencing), as well as fragment analysis. The work of the Laboratory of Genetic Engineering is devoted to the molecular genetic diagnosis of various hematological diseases. With the help of the Nanophore 5 analyzer, the nucleotide sequence of genes can be determined, violations in which lead to the development of certain hereditary or tumor diseases of the blood.
Genetic research is a multi–step process. First, DNA is isolated from the blood of patients. Then, sample preparation is carried out, consisting of several stages. Depending on the size of the gene under study, this process can take from a week up to a month. Then the prepared samples are loaded into Nanophore 5 (up to 96 samples can be analyzed simultaneously). The principle of operation of the analyzer is based on capillary electrophoresis with detection of laser-induced fluorescence (via 7 channels). Special software makes it possible to visualize the results obtained and compare them with reference DNA sequences. This makes it possible to identify the presence or absence of pathogenic variants in the studied genes. The results are given to the clinic in the form of conclusions and used for further scientific research.
In total, there are three similar sequencers at the NMRC for Hematology, which perform diagnostic and scientific research on a daily basis.
